These results suggest a rather neutral effect of trans-11 18:1/CLA butter towards the risk of atherogenesis, whereas trans-10 18:1 butter would tend to be detrimental. In these animals, the VLDL + LDL to HDL ratio was 1.7–2.3 times higher in the trans-10 18:1 group than in the other groups (P = 0.076). Interestingly, trans-9, trans-11 CLA altered the milk fat desaturase index, further demonstrating. In the 12-week rabbits, a shift towards denser LDL, considered as more atherogenic, was shown only in the trans-10 18:1 group. When all treatments were considered, results suggested that trans9, cis-11 was the CLA isomer in the 9, 11 CLA mix responsible for the reduction in milk fat synthesis, although the magnitude was less than that observed for trans-10, cis-12 CLA. Mainly in the 12-week rabbits, higher plasma triglycerides and apolipoprotein B levels shown in the standard and trans-10 18:1 groups compared with those in the trans-11 18:1/CLA group are associated with higher plasma levels of very low density lipoproteins (VLDL) and low density lipoproteins (LDL) also shown in these two groups. Lipid classes were determined enzymatically and apolipoproteins A-I and B by radial immunodiffusion. Lipoproteins were separated by gradient-density ultracentrifugation. Blood samples were collected at the end of dietary treatments. Since the identification of CLA as a factor that can inhibit mutagenesis and carcinogenesis, thousands of studies have been conducted in the last several. New Zealand White male rabbits (9.6 weeks 2.1 kg) were assigned for 6 or 12 weeks to three diets (n = 6 per diet) made of conventional pellets with 0.2% cholesterol and with 12% fat provided from a butter poor in trans-10 and trans-11 18:1 and in CLA (standard group), or rich in trans-10 18:1 (trans-10 18:1 group) or rich in trans-11 18:1 and in cis-9,trans-11 CLA (trans-11 18:1/CLA group). The experiment was designed to study the effects of butters differing in conjugated linoleic acid (CLA) and trans 18:1 contents on lipoproteins associated with the risk of atherogenesis. Cis-9, trans-11-CLA does not promote this detrimental effect.Butters Varying in trans 18:1 and cis-9,trans-11 Conjugated Linoleic Acid Modify Plasma Lipoproteins in the Hypercholesterolemic Rabbit Butters Varying in trans 18:1 and cis-9,trans-11 Conjugated Linoleic Acid Modify Plasma.īauchart, Dominique Roy, Alexandre Lorenz, Stephanie Chardigny, Jean-Michel Ferlay, Anne Gruffat, Dominique Sébédio, Jean-Louis Chilliard, Yves Durand, Denys These results indicate that trans-10, cis-12-CLA intake modifies HDL to form a proatherogenic apoA-II containing particle and promotes phenotypic changes compatible with metabolic syndrome. Significant, positive associations were found between apoA-II and C-III (r=0.883, P<0.001) and between apoA-II and atherosclerosis (r=0.68, P<0.001). The plasma apoA-II concentration increased with its corresponding mRNA concentration in the liver, and was preferentially bound to HDL in the trans-10, cis-12-CLA mice, thus explaining the increased HDL cholesterol concentrations in this group. These reductions were associated with decreased hepatic expression of these proteins and a shift toward lipid-poor apolipoprotein particles. Plasma apoA-I and paraoxonase concentrations were significantly lower in the trans-10, cis-12-CLA group than in the cis-9, trans-11-CLA group. Compared with the control and cis-9, trans-11-CLA mice, those fed the trans-10, cis-12-CLA diet had significantly higher HDL cholesterol concentrations, and had a higher incidence of hypertriglyceridemia and hepatic steatosis.

Most of them use mixtures containing several CLA isomers, mainly cis-9,trans-11 and trans-10, cis-12 in equal concentration. Previous studies have shown that brown adipose tissue (BAT) is particularly sensitive to CLA-supplemented diet feeding.

To test this hypothesis, apoE-deficient mice were fed a Western-type diet enriched with linoleic acid (control), cis-9, trans-11-CLA or trans-10, cis-12-CLA (1.0% wt/wt) for 12 wk, and the effects on HDL metabolism and apoC-III levels recorded. Conjugated linoleic acid (CLA) is reported to have health benefits, including reduction of body fat. This suggests that under these conditions, the HDL apolipoproteins (apo) produced are abnormal. Trans-10, cis-12-conjugated linoleic acid (CLA)-enriched diets promote atherosclerosis in mice despite increasing blood concentrations of HDL cholesterol.
